Northeast Structural Genomics Consortium (NESG)
The neSG Consortium supports academic scientists in 12 research and teaching institutions, with activities in Structural Bioinformatics, Protein Sample Production, X-ray Crystallography, and NMR Spectroscopy. Target selection is coordinated with three other Large-Scale Protein Structure Production Centers of the PSI2 through regular conference calls and meetings. 3D structures are generated primarily from three classes of protein targets: (i) representatives from large protein domain families, (ii) proteins selected from networks associated with human cancer and developmental biology, and (iii) targets nominated by the biomedical community. Specific biochemical functions and/or roles in biological pathways have been discovered based on insights provided by these 3D protein structures. The resulting structure-function hypotheses are explored through collaborations with other research groups, and in follow up projects that are supported by other funding sources.
The NESG project also includes substantial efforts in tracking and disseminating extensive structural genomics data and reagents to the biological community. In addition, the program supports training activities for postdoctoral, graduate, and undergraduate students. Technologies that have been developed include robotic methods of protein sample preparation, single protein production (SPP) systems for isotope enrichment, high throughput biophysical methods for construct optimization, G-matrix Fourier transform (GFT) methods for rapid NMR data collection, and software for automated NMR data analysis. Other key technologies, such as high-throughput crystallization screening, are supported by collaborations with other PSI2 Technology Development Centers. These protein structures, reagents, and technologies are available to the broader scientific community, providing infrastructure that will impact a wide range of applications in biological and biomedical research.
